Molecular glue degraders of HuR suppress BRAF-mutant colorectal cancer

Abstract BRAF gain-of-function mutations, particularly BRAF(V600E), affect roughly 10% of all patients with colorectal cancer (CRC), and portend poor prognosis with limited therapeutic interventions. BRAF inhibitors such as encorafenib are ineffective due to MAPK pathway reactivation driven by BRAF dimerization. Combined inhibition of BRAF and EGFR, although approved therapies, results in short survival benefits and frequent treatment resistance and relapse1,2,3.

A 10 year old Xeon is all you need (for 26B-A4B MTP Drafters without GPU)

A 10 year old Xeon is all you need 17 minutes read The previous post covered getting Gemma 4’s MTP drafters quantized and paired with a verifier. This one is about running the result on a machine that has no business running it. I have a recycled server.

$\mathrm{ECI}_{\mathrm{sem}}$: Semantic Residual Effective Contrastive Information for Evaluating Hard Negatives

arXiv:2603.20990v3 Announce Type: replace Abstract: Hard-negative source selection for dense retrieval is usually decided only after fine-tuning and downstream evaluation. We propose $\mathrm{ECI}_{\mathrm{sem}}$, a semantic residual variant of Effective Contrastive Information (ECI) that ranks candidate negative sources using frozen target-encoder embeddings. $\mathrm{ECI}_{\mathrm{sem}}$ is training-free, not label-free: each scored example requires a query, a labeled positive, and an...

Training-Free Lexical-Dense Fusion for Conversational-Memory Retrieval

Announce Type: new Abstract: Retrieving the few past turns that answer a new query across long multi-session histories is the retrieval bottleneck behind long-term conversational memory (LoCoMo, LongMemEval). Recent concurrent work, Nano-Memory, shows that scoring a session by the maximum query-turn similarity (late interaction, "Turn Isolation Retrieval") beats mean-pooled session embeddings. We do not claim that effect; we replicate it and ask what a training-free, CPU-only retrieval stage...

ECI: Effective Contrastive Information to Evaluate Hard-Negatives

arXiv:2603.20990v2 Announce Type: replace Abstract: Hard-negative source selection for dense retrieval is usually decided only after fine-tuning and downstream evaluation. We propose Effective Contrastive Information (ECI), a training-free diagnostic that ranks candidate negative sources using frozen target-encoder embeddings. ECI is training-free, not label-free: each scored example requires a query, a labeled positive, and an explicit candidate negative.

Mutation-dependent responses to sleep and exercise in clonal haematopoiesis

Abstract Clonal haematopoiesis (CH) activates inflammation and increases the risk of atherosclerosis1,2. Whether lifestyle alters CH clone expansion or the phenotypic programming of CH mutant cells, thereby affecting atherosclerosis, is unknown. Here, in humans and mice and across mutations in Jak2, Tet2, Trp53 and Dnmt3a, we demonstrate mutation-dependent responses to sleep and exercise in CH and show that mutant cells are uniquely sensitive to lifestyle.
