The Greenwith man was rushed to hospital, but died shortly after.
Police said emergency services were called to the popular city nightlife area close to 11PM on Saturday night.
Hindley Street was closed while police managed the scene.
Major Crash Investigators also attended to examine the circumstances around the incident.
The street was reopened around 2:15 AM on Sunday morning.
Police are urging anyone who witnessed the crash and who hasn't yet spoken to them to contact Crime Stoppers.
The man's death was the 70th life lost on SA roads this year.
