Emergency services received reports of a crash near the Nerowie and Parwan-Exford road intersections just before 4pm.
Victorian Police said one child died at the scene while six others were injured.
"Despite CPR being performed, one school-aged child died at the scene," a spokesperson said.
"A second child was airlifted with critical injuries. Five more children were taken to hospital with non-life-threatening injuries.
"One woman was taken to hospital for observation."
Ambulance Victoria said its specialist crews were responding to the incident.
"Two specialist Air Ambulance Victoria helicopter crews, two Mobile Intensive Care Ambulance paramedic crews and five Advanced Life Support paramedic crews are currently on scene," a spokesperson said.
"Their priority is with their patients."
An investigation into the crash is ongoing.
