A genetic tool targeting brain GPR75

G-protein-coupled receptor 75 (GPR75) has emerged as an important mediator in diet-induced obesity (DIO) and a promising therapeutic target for anti-obesity drugs. However, the anatomical location of GPR75 in the brain remains unclear, hindering the understanding of GPR75 biology in DIO. Here, we generated a new GPR75-GFP-Ires-Cre knockin mouse strain, in which the Cre expression is driven by the endogenous GPR75 promoter and the GFP is fused with the C-terminal of the GPR75 protein.

CREP: Cis-Regulatory Element Predictor Based on Fine-Tuned Enformer

A substantial fraction of disease-associated genetic variants reside in non-coding regions of the genome, where they act by perturbing cis-regulatory elements (CREs) such as enhancers, promoters, and insulators. While recent sequence-based deep learning models, such as Enformer, accurately predict continuous epigenomic signals from DNA sequence, they do not directly provide discrete and interpretable CRE annotations. Here, we present CREP (Cis-Regulatory Element Predictor), a fine-tuned...

Schwann Cell Mapping and Characterization in Bone of Different Embryonic Origins

Schwann cells (SCs) provide support for nerves throughout the body. Despite importance for nerve function and repair, the morphology and distribution of SCs in bone remains largely undefined. In this study we used a ,Schwann Cell Mapper, mouse (Mpz-Cre+/-;TdT+/+;Ngfr-eGFP+/+) and a ,p75 Lineage Tracer, mouse (Ngfr-CreERT2+/-;ZsGreen1+/+) to study SC localization and morphology within the adult mouse calvaria, limb, and vertebrae.

Biological Reasoning-Informed Regression for Interpretable Regulatory DNA Activity Prediction

arXiv:2606.08147v1 Announce Type: cross Abstract: DNA cis-regulatory elements (CREs) such as enhancers control gene expression levels. Accurately predicting regulatory activity from DNA sequences is valuable but challenging, as it requires understanding complex biological regulatory processes. Existing methods typically regress activity scores from sequences in a black-box manner, limiting both interpretability and regression performance.

'All-in-one' platform developed for multiple trait stacking in crops

'All-in-one' platform developed for multiple trait stacking in crops Gaby Clark Scientific Editor Alexander Pol Deputy Editor A major goal of modern crop breeding is to efficiently combine multiple desirable traits by "stacking" the favorable gene variants (alleles) that contribute to those traits in a single crop variety. However, current strategies are often time-consuming and inefficient. Now, a team led by Gao Caixia at the Institute of Genetics and Developmental Biology of the Chinese...

Cell-type targeted CRISPR/Cas9 Clock knockdown in mouse VTA dopamine neurons alter sleep, behavior, and cellular excitability

Bipolar disorder (BD) is a severe psychiatric disease characterized by recurrent mania, depression, and circadian rhythm disruption. Among circadian regulators implicated in mood-related dysfunction, Clock has emerged as a particularly strong mechanistic candidate. However, cell type-specific functions of Clock within mood-relevant circuits remain incompletely defined.

Hypertension-mediated cardiac fibrosis is a mechanical process initiated by smooth muscle cells

Hypertension represents the most prevalent chronic cardiovascular condition, typically culminating in pathological cardiac remodeling characterized by hypertrophy and extensive fibrosis. Although the cellular phenotypes associated with these changes are well-documented, the precise mechanisms by which hypertensive stress is sensed and transduced into a fibrotic program remain poorly defined. To elucidate these mechanisms, we investigated the role of Lysyl oxidase (LOX), an extracellular...
